Compared with rat and human islets, the expression of GluR1/GluR2 in human beta TC3, RIN-m5F, and HIT cells was significantly lower and expression of GluR3/GluR4 was significantly higher. In the islet cell lines, the GluR1/GluR2 subunit was expressed in INS-1 cells, and the GluR3/GluR4 subunit was expressed in HIT-T15 cells. In the beta TC3, RIN-m5F, and HIT cells, the alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid-evoked inward currents were blocked by 100 micromol/L NBQX, a selective AMPA receptor antagonist. However, in INS-1 and HIT-T15 cells, the alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid-evoked inward currents were not blocked by NBQX.
